Humidity is one of the most important variables during tissue culture acclimation. Plants leaving an in-vitro vessel have developed under extremely humid conditions with limited air movement, so moving them directly into ordinary room humidity can cause rapid water loss before their leaves and roots are ready to regulate it.
For most tissue culture plants, humidity should be reduced gradually rather than dropped all at once. Start with a humid protected environment after deflasking, then progressively increase ventilation as the plant maintains leaf turgor and begins active ex-vitro growth.
Why do tissue culture plants need high humidity after deflasking?
In-vitro plants develop in a very different environment from conventional houseplants. Their leaves have formed at very high relative humidity, while roots have developed in agar or another culture medium rather than an aerated potting substrate. Stomatal control, the cuticle and root water uptake therefore need time to adjust after transfer.
The aim of acclimation is not to keep the plant permanently at extreme humidity. High humidity is a temporary bridge while the plant develops functional ex-vitro roots and becomes better able to control water loss.
What humidity should tissue culture plants start at?
There is no single RH percentage that is correct for every species, plant size or growing system. A useful practical approach is to begin in a covered propagator or similarly humid enclosure where freshly deflasked plants remain turgid, then use plant response rather than an arbitrary calendar alone to determine when to increase ventilation.
Very small plantlets, plants with little functional root mass and thin-leaved species generally need a more cautious transition than robust plantlets with substantial roots.
How fast should humidity be reduced?
Reduce humidity in stages. During the first stage, keep the enclosure mostly closed while watching for persistent wilting, collapse or excessive condensation. Once plants remain firm and stable, introduce a small amount of ventilation. Increase ventilation progressively over subsequent days as long as the plants continue to hold their leaves without prolonged wilting.
If plants wilt strongly after ventilation is increased, return to the previous successful level rather than immediately exposing them to still drier air. Acclimation is a physiological transition, not simply a fixed number of days under a lid.
Ventilation matters as much as the RH number
A sealed propagator can maintain high humidity, but stagnant saturated air can also keep foliage and substrate surfaces wet. Gradually opening vents or increasing the lid gap changes both humidity and air exchange. This makes ventilation a practical way to harden plantlets while reducing the risk of keeping the rooting zone continuously saturated.
How do you know a tissue culture plant is ready for lower humidity?
Useful signs include leaves remaining turgid after ventilation increases, new leaf expansion, new root activity and reduced dependence on the closed propagator. New growth produced after deflasking is particularly useful because it develops under the new environment rather than the original culture vessel conditions.
What if the plant wilts when the lid is opened?
Short-term loss of turgor can indicate that transpiration has increased faster than the plant can replace water. Increase humidity again or reduce the ventilation step, then retry more gradually. Avoid responding by saturating the substrate: a plant can wilt because of poor water balance while its root zone is already too wet.
High humidity does not mean waterlogged substrate
Atmospheric humidity and substrate moisture are separate variables. During acclimation, the plant may benefit from humid air while its roots still require oxygen. A fine, aerated substrate that remains evenly moist without becoming stagnant is usually more useful than a dense medium kept continuously waterlogged.

Light and temperature during humidity reduction
Freshly deflasked plantlets should not be pushed with intense light while their water regulation is still developing. Higher light, temperature and air movement can all increase water loss. Keep conditions stable during the early transition and increase environmental stress gradually as the plant establishes.
Common acclimation mistake: removing the lid too quickly
A plant can look healthy while enclosed and then collapse rapidly when moved straight into ordinary room humidity. This does not necessarily mean the roots have failed. The change in vapour pressure and transpiration demand can simply be too large for the plant at that stage.
A staged transition gives the plant time to produce leaves and roots better adapted to ex-vitro conditions.
Common acclimation mistake: keeping plants sealed for too long
The opposite problem is leaving established plantlets permanently sealed because they look healthy at high humidity. Once plants are actively growing, progressive ventilation is necessary if the goal is to establish them under normal growing conditions.
A practical tissue culture humidity sequence
- Deflask and clean the plantlets carefully.
- Plant into a moist, aerated acclimation substrate.
- Place them in a humid protected enclosure under moderate light.
- Allow the plants to stabilise before making large humidity changes.
- Introduce a small amount of ventilation.
- Increase ventilation gradually while monitoring leaf turgor and new growth.
- If persistent wilting occurs, return temporarily to the previous successful stage.
- Continue until the plants remain stable with the enclosure fully ventilated or removed.
How long does tissue culture acclimation take?
The useful answer is: until the plant is physiologically established outside culture. Species, plantlet size, root condition, substrate, temperature, light and humidity all affect the rate. Treat a timetable as a guide and the plant's response as the deciding signal.
