Do not repot a tissue culture plant simply because a fixed number of days has passed. The safest time is when the plantlet has completed the most vulnerable part of acclimation, is producing stable new growth and has developed an active root system that holds its first substrate together. Repotting too early adds another major change while the plant is still adapting from in-vitro conditions.

Why newly acclimated tissue culture plants should not be repotted too quickly
A plantlet leaving culture is simultaneously adapting its leaves, water relations and roots to a much less controlled environment. Its in-vitro roots may not initially function like roots produced after acclimation, while new ex-vitro roots are still developing.
Moving the plant again can break fine roots, alter moisture and aeration around them and temporarily reduce water uptake. A recently deflasked plant therefore gains little from an unnecessary early repot unless its original container or substrate is causing a genuine problem.
Signs a tissue culture plant is ready to repot
Look for several signs together rather than relying on one measurement. A well-established plantlet should normally be growing rather than merely surviving.
- New leaves: it has produced healthy growth under ex-vitro conditions.
- Active roots: new roots are visible at the side or base of a transparent pot, plug or propagation cell, or the root system has begun to occupy the substrate.
- Substrate stability: the roots hold at least part of the rooting medium together when handled carefully.
- Stable hydration: the plant remains firm under its normal acclimation humidity rather than rapidly collapsing whenever humidity falls slightly.
- No active rot: the stem, crown and roots are healthy enough that repotting is not being used to disguise an unresolved disease or moisture problem.

New growth is more useful than the age of the plantlet
The original leaves were formed in culture. New leaves produced during acclimation provide better evidence that the plant is functioning in its new environment. This is why a calendar rule such as “repot after two weeks” is less useful than observing growth and root development.
Species, plantlet size, temperature, light, substrate, humidity and the condition of the plant when deflasked can all change the rate of establishment.
How much root growth is enough?
You do not need to wait until a small pot is completely root-bound. The aim is to have enough active root growth that the plant can tolerate handling and the first substrate can be moved without the root system falling apart.

Should you remove the original acclimation substrate?
Usually there is no advantage in stripping healthy substrate from established new roots. If the first medium is clean, structurally suitable and not waterlogged, preserve as much of the functioning root zone as practical and pot around it with the next substrate.

Choosing the next pot size
Move up gradually. A very small root system placed into a disproportionately large volume of wet substrate can remain surrounded by moisture for too long. Choose a pot that accommodates the existing roots with some room for expansion without creating a large unused wet zone.
Drainage and substrate structure matter more than simply choosing a particular pot diameter.
What substrate should you use after acclimation?
Once an aroid plantlet is established, the next mix can become closer to the substrate used for a juvenile houseplant, but it should still provide good root-zone aeration. Avoid making an abrupt transition from a fine, evenly moist acclimation medium into an extremely coarse mix that dries around a small root system too quickly.
The transition can be gradual: maintain enough water availability for the small root system while increasing structure as the plant grows.
Should humidity be changed at the same time as repotting?
Avoid stacking several environmental changes together where possible. If a plant has only just been moved to substantially lower humidity, allow it to stabilise before repotting. Likewise, after repotting, keep the surrounding conditions reasonably consistent while disturbed roots recover.

When should you repot early?
Occasionally intervention is necessary before ideal establishment. Examples include a collapsing substrate, persistent waterlogging, severe compaction, contamination spreading through the medium or a container that is physically unsuitable for drainage.
In these situations, the risk of leaving the plant in a damaging root environment can outweigh the risk of disturbance. Handle the roots as little as possible and correct the underlying problem rather than repeatedly repotting.
Repotting after separating multiple plantlets
If several plantlets were grown together in one culture vessel, separating them already creates substantial root disturbance. It is generally better to place each separated plantlet directly into an appropriately sized first acclimation container rather than repeatedly moving it.

After repotting
Water the new substrate evenly enough to settle it around the existing root zone, but do not keep it saturated simply because the plant came from tissue culture. Maintain stable light and temperature and watch the plant rather than immediately changing several other variables.
A short pause in visible growth after root disturbance can occur. Continued collapse, spreading stem softness, persistent substrate saturation or progressive root loss suggests a problem that should be investigated rather than treated as normal transplant shock.
